No, I've already unlocked Dry Bones and Daisy, and I'm still gettin my ass handed to me in the online competitions. I've also seen a few people who have unlocked the R.O.B. character, which seems like an impossible feat to me since I'm still trying to beat the 100cc Special Cup in single player mode.
